Ege \u00dcniversitesi Rekt\u00f6r\u00fc Prof. Dr. Necdet Budak, \u00fcretken bilim insan\u0131 Dr. \u00d6\u011fretim \u00dcyesi Yasin Kaymaz\u2019\u0131 tebrik ederek ba\u015far\u0131lar diledi.<\/p>\n<p style=\"text-align: justify;\">Ara\u015ft\u0131rma ile ilgili bilgi veren Dr. \u00d6\u011fretim \u00dcyesi Yasin Kaymaz,\u00a0 \u201cTek h\u00fccre RNA sekanslama y\u00f6ntemleri genellikle transkripte olan genlerin 3\u2019-UTR ucunun yakalanarak sekanslanmas\u0131 prensibine dayanmaktad\u0131r. Bu sayede elde edilen sekans okumalar\u0131 genin ekspresyon seviyesini \u00f6l\u00e7mede kullan\u0131l\u0131rken potansiyel olarak genin hangi transkript izoformunun daha aktif oldu\u011fu bilgisini de verebilmektedir. Buna ek olarak KHDAK t\u00fcm\u00f6r tek h\u00fccre transkriptom profilleri bug\u00fcne kadar t\u00fcm\u00f6re \u00f6zg\u00fc PA noktalar\u0131n\u0131n incelenmesine konu olmam\u0131\u015ft\u0131r. Bu bak\u0131\u015f a\u00e7\u0131s\u0131 ile tek h\u00fccre RNA sekanslama verilerinin tekrar biyoinformatik incelemeden ge\u00e7irilmesi gerekmektedir\u201d dedi.<\/p>\n<p style=\"text-align: justify;\">Dr. \u00d6\u011fretim \u00dcyesi Yasin Kaymaz,\u00a0 \u201cTek h\u00fccre sekanslama \u00e7al\u0131\u015fmalar\u0131yla olu\u015fan birden fazla veri setinin entegrasyonu ile t\u00fcm\u00f6r h\u00fccrelerinin normal h\u00fccrelerle kar\u015f\u0131la\u015ft\u0131rmal\u0131 analizi sonucu poliadenilasyon noktalar\u0131n\u0131n belirlenmesi ve t\u00fcm\u00f6re \u00f6zg\u00fc profillerinin \u00e7\u0131kar\u0131lmas\u0131 hen\u00fcz KHDAK i\u00e7in ger\u00e7ekle\u015ftirilmemi\u015ftir. Bu proje ile akci\u011fer kanseri tek h\u00fccre ara\u015ft\u0131rmalar\u0131nda mevcut olan bu eksiklik giderilmeye \u00e7al\u0131\u015f\u0131lacakt\u0131r. Tek h\u00fccre t\u00fcm\u00f6r transkriptom profilleri baz\u0131nda alternatif poliadenilasyon kullan\u0131m\u0131n\u0131n hasta prognostik verileri ile ili\u015fkilendirilmesini i\u00e7eren bir \u00e7al\u0131\u015fmaya hen\u00fcz literat\u00fcrde rastlanmam\u0131\u015ft\u0131r. Bu \u00e7al\u0131\u015fma ile alternatif poliadenilasyon kullan\u0131m\u0131n\u0131n t\u00fcm\u00f6r prognostik veriler ile ili\u015fkisi incelenerek uzun d\u00f6nemli hayatta kal\u0131ma (long-term survival) etkisi incelenecektir\u201d diye konu\u015ftu.<\/p>\n<p style=\"text-align: justify;\"><b>T\u00fcm\u00f6r h\u00fccrelerinin profilleri belirlenecek<\/b><\/p>\n<p style=\"text-align: justify;\">Projenin amac\u0131 ile ilgili detaylar\u0131 da payla\u015fan Dr. \u00d6\u011fretim \u00dcyesi Yasin Kaymaz,\u00a0 \u201cBu proje ile \u00f6nceden \u00fcretmi\u015f oldu\u011fumuz \u2018endSeqTools\u2019 isimli biyoinformatik arac\u0131m\u0131z\u0131 APA takaslar\u0131 i\u00e7in istatistik testleri de yapabilir \u015fekilde tek h\u00fccre sekanslama verilerine de uyarlayarak yeni bir yakla\u015f\u0131m ger\u00e7ekle\u015ftirecektir. Tek h\u00fccre sekanslama verileri h\u00fccre t\u00fcr\u00fcne \u00f6zg\u00fc alternatif poliadenilasyon noktas\u0131 profillerinin \u00e7\u0131kar\u0131lmas\u0131na ve bu profillerin fonksiyonel olarak \u00e7al\u0131\u015f\u0131labilmesine olanak sa\u011flayabilir. Bu ara\u015ft\u0131rman\u0131n temel amac\u0131 tek h\u00fccre transkriptom verileri kullan\u0131larak normal h\u00fccrelerden farkl\u0131 olarak KHDAK t\u00fcm\u00f6r h\u00fccrelerine \u00f6zg\u00fc 3\u2019-UTR kullan\u0131m\u0131 ve alternatif poliadenilasyon noktalar\u0131n\u0131n belirlenmesidir. Bu ama\u00e7la \u00f6nceden yay\u0131nlanm\u0131\u015f \u00e7al\u0131\u015fmalardaki akci\u011fer kanseri tek h\u00fccre RNAseq verileri elde edilecek ve bu ama\u00e7 i\u00e7in yeniden dizayn edece\u011fimiz biyoinformatik arac\u0131m\u0131z ile t\u00fcm\u00f6r h\u00fccrelerinin poliadenilasyon profilleri belirlenecektir.
