Bu farkl\u0131las\u0327m\u0131s\u0327 rolleri tetikleyen veya fonksiyonlar aras\u0131 ge\u00e7is\u0327genlig\u0306i sa\u011flayan temel molek\u00fcler mekanizmalar hen\u00fcz tamamen a\u00e7\u0131kl\u0131k kazanmam\u0131\u015ft\u0131r. Makrofajlar\u0131n fonksiyonel olarak \u00f6zelles\u0327meleri ile micro RNAlar\u0131n ilis\u0327kili bulunmas\u0131 3\u2019-UTR \u00fczerinden reg\u00fclasyonlar\u0131n varl\u0131g\u0306\u0131n\u0131 is\u0327aret etmekte ve alternatif poliadenilasyon ile olus\u0327an transkript izoformlar\u0131n\u0131n tespit edilmesinin \u00f6nemi vurgulamaktad\u0131r. Tek h\u00fccre RNA sekanslama teknolojilerinin ilerlemesi ile birlikle bu ili\u015fkilerin molek\u00fcler nedenleri doku g\u00fcr\u00fclt\u00fcs\u00fcnden uzakla\u015fm\u0131\u015f\u0327, benzer genetik programa sahip h\u00fccre t\u00fcrleri \u00fczerinde ara\u015ft\u0131r\u0131lma imk\u00e2n\u0131 bulmaktad\u0131r. Global transkriptom \u00f6l\u00e7ekte \u00f6zelles\u0327mis\u0327 makrofaj alt gruplar\u0131n\u0131n UTR uzunlu\u011fu de\u011fi\u015fimleri ile potansiyel mikro RNA temelli reg\u00fclasyonlar aras\u0131nda ili\u015fki kurulmas\u0131 a\u00e7\u0131s\u0131 \u00e7al\u0131\u015fman\u0131n bir di\u011fer \u00f6zg\u00fcn de\u011feridir. Ayr\u0131ca makrofaj polarizasyonu akabinde \u00fcretilecek tek h\u00fccre RNA sekans verileri TAM\u2019lar\u0131n ayd\u0131nlat\u0131lmas\u0131nda bir referans modeli olarak kullan\u0131lacak olmas\u0131 projenin \u00f6nemli bir \u00f6zg\u00fcn yakla\u015f\u0131m\u0131d\u0131r. Bu proje ile t\u00fcm\u00f6rle ili\u015fkili makrofajlar\u0131n alt gruplar\u0131n\u0131n biyomark\u00f6rleri olma potansiyelinde transkriptomik farkl\u0131l\u0131klar\u0131n tek h\u00fccre sekanslama verileri ile elde edilme imk\u00e2n\u0131 sag\u0306lanmaktad\u0131r. Biyoinformatik algoritma geli\u015ftirme s\u00fcreci, kontroll\u00fc deney ile referans verisi \u00fcretimi, h\u00fccre alt gruplar\u0131n\u0131 belirleyen model geli\u015ftirilmesi ve geli\u015ftirilen modelin t\u00fcm\u00f6r biyopsi \u00f6rne\u011finde test edilmesi seklinde 4 ana safhadan olu\u015fan proje 9 is\u0327 paketi i\u00e7ermektedir. 24 ay i\u00e7inde tamamlanmas\u0131 planlanan bu proje \u00e7e\u015fitli a\u015famalar\u0131nda Ege \u00dcniversitesi, Hacettepe \u00dcniversitesi ve Antalya Bilim \u00dcniversitesi is\u0327 birlig\u0306i sayesinde ger\u00e7ekle\u015ftirilecek olup olduk\u00e7a interdisipliner alanlar\u0131n (biyoinformatik, bilgisayar m\u00fchendislig\u0306i, imm\u00fcnoloji, molek\u00fcler onkoloji) etkile\u015fimi ve akademik \u00fcretkenlig\u0306ini art\u0131racakt\u0131r. Son teknoloji molek\u00fcler tekniklerden tek h\u00fccre RNA sekanslama y\u00f6ntemlerinin en d\u00fcs\u0327\u00fck maliyetli ve y\u00fcksek verimlilikte bir metodunu \u00fclkemizde ilk defa uygulama f\u0131rsat\u0131 verecek bu proje bu y\u00f6n\u00fcyle de teknoloji transferi imk\u00e2n\u0131 sunma potansiyeliyle ve \u00fclkemize hem ekonomik hem de akademik a\u00e7\u0131dan b\u00fcy\u00fck katk\u0131lar sa\u011flayacakt\u0131r.
